How to read this
- AmphaFluid buffer = recommended AmphaFluid for pollen viability measurement.
- Alt. = alternative AmphaFluid to try for other purposes (development stages, ploidy).
- ★ Crop-specific AmphaChip available for this crop — an optional dedicated chip for the Ampha P20 with automated data analysis. The standard chip works too.
- Chip & Filter size must match your pollen size. Pollen size can vary ±20% or more; polyploidy increases size.
- Templates are the AmphaSoft PRO measurement templates — contact support for the files.
Sample prep tips
- Dried / stored pollen: rehydrate and equilibrate in buffer before measuring.
- Sticky or oily pollen: add 0.05% Tween 20 or 80.
- Microspores / protoplasts: most species work with AmphaFluid 6, or dilute the isolation medium with AmphaFluid 6 (1:4 to 1:6).
- Use fresh samples where possible, filter before use, and measure immediately.
